Cas9 and sgRNA Structure
The constant part of the sgRNA directing Streptococcus pyogenes Cas9 (SpCas9) consists of a fused crRNA and tracrRNA and adopts a characteristic folding pattern important for Cas9 activity.
PlatinumCRISPr embeds each candidate guide into this scaffold and evaluates whether the resulting RNA retains essential structural elements while correctly pairing with the DNA target.
